When you stop and fill up for gas, be sure the doors and windows are securely shut and let her out for a few minutes of playtime and cuddles. Offer her some food and water and then put her in a litterbox to see if she uses it. If she is a good lap kitten and tolerates holding, you could have the non-driver sit by the carrier and hold her securely in a lap for a while. But if she gets restless she needs to go back into the carrier.
For her safety and for yours the kitten needs to be restrained to the carrier while the car is in motion. And the carrier needs to be restrained to the seat.
